BackgroundMonkeypox is a rare zoonosis caused by monkeypox virus, which has become the most serious orthpoxvirus and consists of complex double stranded DNA. The cases are mostly in central and western Africa. The pathogenesis of monkeypox is that the virus invades the body from respiratory mucosa, multiplies in lymphocytes, and incurs into blood producing transient venereal toxemia. after the virus multiplies in cells, the cells can invade the blood and propagate to the skin of the whole body, causing lesions.C19L is palmytilated EEV membrane glycoprotein；similar to vaccinia virus strain Copenhagen F13L.The poxviridae are enveloped unsegmented dsDNA viruses.Unlike many dsDNA viruses that replicate in the host nucleus poxviruses replicate in the cytoplasm, viral genes are expressed in a bi-phasic manner with early genes encoding non-structural proteins involved in genome replication and late genes encoding the viral structural proteins.

Expression systemC19L, produced in E.coli is a single non-glycosylated polypeptide chain containing 372 amino acids. A fully biologically active molecule has a molecular mass of 41.8kDa. The apparent molecular mass of the protein is approximately 42 kDa analyzed by reducing SDS-PAGE and is obtained by chromatographic techniques.
